Abstract:
Objective: To establish polymerase chain
reaction (PCR) using DNA of M. leprae from fixed and paraffin-embedded
tissue(FPET). Methods: The DNA of M.
leprae to be released from FPET and purified by using
Texpat Kit and 100% alcohol, then the PCR was conducted.
Results: 32 samples, BI of which were
all more than 1+, 28/32 positive reactions were yielded,
1 sample, its BI was equal to zero, negative result was
yielded. The sensitivity of PCR reached 0.04pg of DNA
level. Conclusion: This method is very
useful to amplify the DNA of M. leprae from fixed and
paraffin embedded tissue.
